Protein expression analysis

from clipboard

Strain specific Apo(a) protein expression analysis in B-hLPA mice by ELISA. Serum was collected from wild-type C57BL/6 mice (+/+), heterozygous B-hLPA mice (H/+), homozygous B-hLPA mice (H/H), all mice in 7-week-old. 3 B-hLPA mice of each gender for detection, 1 wild-type C57BL/6 mice of each gender as a control. Human Apo(a) was exclusively detectable in heterozygous and homozygous B-hLPA mice, but not in wild-type C57BL/6 mice.

Effect of Olpasiran analog on plasma apo(a) in B-hLPA mice

Dosing regimen

from clipboard

Changes of plasma apo(a) levels in heterozygous B-hLPA mice in response to olpasiran analog. Olpasiran analog (provided by the client) is a double-stranded small interfering RNA (siRNA) targeting lipoprotein(a) and potently reduce plasma Lp(a) concentration. As shown in the panel, Olpasiran analog reduced Apo(a) concentrations in heterozygous B-hLPA mice(A), achieving up to over 80% reduction from baseline(B) after administration of a single dose. The mice can be used for preclinical studies of efficacy.
