Description 

This B-Luc CT26.WT cell line expresses firefly luciferase as a marker of CT26.WT cells. Fluorescence could be detected in B-Luc CT26.WT cells.

Application

B-Luc CT26.WT cell line can be used to evaluate anti-cancer drugs in colon cancer in vivo. It can be applied subcutaneously and orthotopically to colons in mice

Targeting strategy

Gene targeting strategy for B-Luc CT26.WT cells. The CMV promoter and luciferase were inserted into the targeting vector used in the lentivirus system.

Luminescence signal intensity of B-Luc CT26.WT cells. Luminescence intensity was measured using the Bright-GloTM luciferase Assay (Promega, Cat E2610). B-Luc CT26.WT cells have a strong luminescence signal. The mix clone of B-Luc CT26.WT cells can be used for in vivo experiments.

B-Luc CT26.WT cells (1x105, 5x105) were injected into the colon of wild-type BALB/c mice. Signal intensity and body weight were measured twice a week. (A) Imaging was performed twice a week. (B) Body weight (Mean ± SEM). B-Luc CT26.WT cells can be used for in vivo efficacy evaluation.

Subcutaneous homograft tumor growth of B-Luc CT26.WT cells. B-Luc CT26.WT cells (1x105) and wild-type CT26.WT cells (1x105) were subcutaneously implanted into BALB/c mice (female, 9-week-old, n=6). Tumor volume and body weight were measured twice a week. (A) Average tumor volume ± SEM. (B)  Body weight (Mean± SEM). Volume was expressed in mm3 using the formula: V=0.5 X long diameter X short diameter2. As shown in panel A, B-Luc CT26.WT cells were able to establish tumors in vivo and can be used for efficacy studies.
