Generation of gene editing cell line and expression analysis
Gene targeting strategy for B-luc EL4. The luciferase sequence was inserted in the ROSA26 exon1 of wild-type EL4.
Phenotypic Analysis
The luminescence signal intensity of B-luc EL4 cells. The luminescence intensity was detected by Bright-GloTM luciferase Assay System (Promega, Cat E2610), and B-luc EL4 cells have strong luminescence signal.
Imaging analysis in vivo
In vivo Imaging Analysis
Tumor growth and in vivo imaging of B-luc EL4 cells. B-luc EL4 cells (2x105) were injected by tail vein into C57BL/6 mice (female, 7-8 week-old, n=5). Signal intensity and body weight were measured twice a week. (A) Imaging was performed on days 0, 3, 6, 10, 13, 17, 20, 24, and 27, (B) Mice body weight (Mean ± SEM). B-luc EL4 cells can be used for in vivo efficacy evaluation.
