Protein expression analysis
Strain specific TLR2 expression analysis in heterozygous B-hTLR2 mice by flow cytometry. Peritoneal exudative macrophages(PEMs) were collected from wild-type C57BL/6 mice(+/+) and heterozygous B-hLTR2 mice(H/+). Mouse TLR2 was detectable in the WT and heterozygous B-hTLR2 mice, Human TLR2 was exclusively detectable in the heterozygous B-hTLR2 mice.
mRNA expression analysis
Strain specific analysis of TLR2 gene expression in WT and hTLR2 mice by RT-qPCR. Spleens were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hLTR2 mice (H/H). The mRNA expression of hTLR2 in B-hTLR2 mice (H/H) was less than those in the C57BL/6 mice (+/+), demonstrating that introduction of hTLR2 in place of its mouse counterpart changes the mRNA expression level of TLR2.
