Gene Description


KLB(Klotho Beta) ,also known as BKL, is expressed primarily in the liver and pancreas, with lower expression in adipose tissue. Like Klotho, Klotho beta facilitates binding between FGF19 subfamily members and their receptors via the formation of a ternary complex. The Klotho beta mediated interaction of FGF15 (human FGF19) with FGF Receptor 4 in the liver negatively regulates bile acid synthesis by controlling the secretion of two key bile acid synthase genes, cholesterol 7-alpha hydroxylase (Cyp7a1) and sterol 12-alpha hydroxylase (Cyp8b1). Klotho beta is also a cofactor for the interaction of FGF21 with FGF Receptor 1c in adipocytes, which allows FGF21 to stimulate GLUT1 expression, upregulating adipocyte insulin-dependent glucose uptake. Diseases associated with KLB include outlet dysfunction constipation and functional diarrhea. Gene ontology (GO) annotations related to this gene include hydrolase activity, hydrolyzing O-glycosyl compounds, and fibroblast growth factor binding. An important paralog of this gene is KL.

Strain specific analysis of KLB expression in heterozygous B-hKLB  mice by Western blot. 
Mouse KLB was detectable both in liver of wild type (+/+) mice and heterozygous B-hKLB  mice. Human KLB was detectable in heterozygous B-hKLB  mice (H/+) , HepG2 cells, and Hep3B cells but not in +/+ mice. HepG2 and Hep3B cells were used as positive control there.
