Gene description


CD70 (CD70 molecule, gene ID 970), also known as tumor necrosis factor ligand superfamily member 7 (TNFSF7), CD27 ligand (CD27-L, CD27L, CD27LG), and KI-24 antigen, encodes a type II transmembrane glycoprotein. CD70 is a member of the TNF superfamily and the ligand for the T cell costimulatory receptor CD27. The interaction of CD70 with its receptor CD27 promotes expansion and differentiation of memory and effector T cells as well as B-cell expansion and plasma cell differentiation. In normal tissues, CD70 expression is restricted to cells of hematopoietic origin, and it is only transiently induced upon activation of T and B lymphocytes, natural killer cells, and maturation of dendritic cells. However, aberrant CD70 expression was found in hematologic malignancies and also in solid tumors, such as renal cell carcinoma, leukemia/lymphoma, NSCLC, and melanoma. CD70 expression on malignant cells coupled with its highly restricted expression on normal cells makes CD70 an attractive target for antibody based therapies. Clinical trials evaluating CD70 as a therapeutic target include the use of antibody–drug conjugates, enhanced antibody-dependent cellular cytotoxicity with an anti-CD70 monoclonal antibody, and CAR-T cell therapy.

Strain specific analysis of CD70 gene expression in wild type (WT) mice and B-hCD70 mice by RT-PCR. Mouse CD70 mRNA was detectable only in thymocytes of WT mice (+/+) and B-hCD70 mice (H/+). Human CD70 mRNA was detectable only in homozygous B-hCD70 mice (H/+) but not in WT mice (+/+).

Strain specific CD70 expression analysis in homozygous B-hCD70 mice by flow cytometry. Bone marrow cells were collected from wild type (WT) mice (+/+) and homozygous B-hCD70 mice (H/H) stimulated with LPS in vivo, and analyzed by flow cytometry with species-specific anti-CD70 antibody. Mouse CD70 was detectable in WT mice (+/+). Human CD70 was exclusively detectable in homozygous B-hCD70 mice (H/H) but not in WT mice (+/+).
